1、二者含義不同。
a number of 表示：許多，一些，相當於a lot of ，some。the number of 表示：……的數量。
2、二者用法不同。
a number of 是「大量的」，等同於 a lot of一類，因此後面接名詞複數，謂語通常用複數。
a number of students were absent from the meeting. 許多學生沒有到會.（were是謂語，用複數）
the number of是「......的數量」，後面接名詞複數，但謂語要用單數。
例：the number of students absent from the meeting was surprising. 沒有到會的學生人數很驚人.（was是謂語，用單數）
3、短語本質不同。
a number of 為量詞短語，表示「許多……」，中心詞是這個短語所修飾的複數名詞。
the number of表示「……的數量」，後接複數名詞，其中，中心詞為 number。
與a number of 和 the number of 類似的易混淆的短語還有如下幾組短語：
1、in front of, in the front of
in front of 範圍外的前面。例：there』s a bus stop in front of the house .
房子前面有一個公共汽車站（在公路的對面，即範圍外的前面。）
in the front of 範圍內的前面。例：in the front of the room sits a boy.
房間裡坐著一個男孩。（房間裡面的前面，就是範圍內的前面）
2、three of us, the three of us
three of us 我們(不止三個)中的三個。例：three of us sweep and mop the floor and the remainder clean the window.
我們之中三人掃地拖地板，其餘的人擦玻璃。
the three of us 我們三個(就三個人)例：the three of us - tom, jack and i went to the cinema.
3、of the day, of a day
of the day 每一天的，當時的，當代的。例：a famous scientist of the day. 一個當代著名的科學家。
of a day 暫時的，不長久的。例：i had a bummer of a day.我這一天都很煩。

the number of 意思是「.的總數」,後接複數名詞,但謂語要用單數：
the number of students absent from the meeting was surprising.
沒有到會的學生人數很驚人.
a number of 意思是「許多...」,後接複數名詞,謂語通常用複數：
